Understanding Semax Dosing in Research
Semax is a synthetic peptide analog of the adrenocorticotropic hormone (ACTH) fragment 4-10 (Met-Glu-His-Phe-Pro-Gly-Pro), developed at the Institute of Molecular Genetics in Russia. It has been extensively researched for its nootropic, neuroprotective, and neurorestorative properties, with particular focus on BDNF modulation and cognitive enhancement.
Mechanism of Action
Semax increases brain-derived neurotrophic factor (BDNF) expression, modulates serotonergic and dopaminergic systems, and enhances NGF (nerve growth factor) levels. Unlike full-length ACTH, Semax lacks steroidogenic activity, making it a cleaner research tool for studying melanocortin-mediated neuroprotection without adrenal axis effects.

Route of Administration Considerations
Intranasal administration provides rapid CNS delivery via the olfactory and trigeminal nerve pathways, bypassing the blood-brain barrier. This route achieves higher CNS bioavailability compared to subcutaneous injection, which is why intranasal doses in research are typically higher (compensating for mucosal absorption losses) while still achieving comparable brain tissue concentrations.
Dose-Response Research
Russian clinical studies demonstrated dose-dependent cognitive enhancement with Semax. At the standard clinical dose (0.1% solution, 3 drops per nostril), significant improvements in attention, memory formation, and cognitive flexibility were observed. Higher concentrations (1% solution) showed enhanced neuroprotective effects in neurological research contexts.
Comparison with Selank
Semax is often studied alongside Selank, another Russian-developed peptide. While Semax primarily modulates BDNF and dopaminergic pathways (nootropic/stimulatory profile), Selank acts primarily through enkephalin metabolism and GABAergic modulation (anxiolytic/calming profile). They are considered complementary in neuroscience research.
Reconstitution
